New Hypotensive and Mitogenic Peptides : Salusin-a and Salusin b

Salusins
Bioinformatic analysis of cDNA and genomic DNA sequence information has rapidly increased the rate of discovery of endogenous peptides of biological interest. In 2003, Shichiri and coworkers reported the latest endogenous ligands to be discovered in this fashion, the salusins. The salusins were translated from an mRNA splice of TOR2A, which is a member of the torsion dystonia family (1).

Hypotension and bradycardia (slowed heart rate) resulted at concentrations of 1 - 10 nmol/kg of salusin-a and 0.1 - 1 nmol/kg of salusin-b upon iv administration to rats. However, only salusin-b was reported to stimulate Arg8-vasopressin release in a dose-dependent manner. It was postulated that salusin-b affects cell proliferation due to increased [Ca2+] via influx of extracellular Ca2+ into cells and induced expression of growth-associated proteins, while salusin-a exhibits the same but to a lesser extent. Both salusins are found in human tissues, plasma, and urine suggesting endocrine process involvement. The Japanese research group believes that the salusins may contribute to the regulation of hemodynamics via systemic cardiovascular routes as well as pituitary endocrine functions. The exact mechanism of action remains unclear. Further studies of these hypotensive and mitogenic peptides should prove fascinating.
